Suitably modified peptides modeled on histone tails were obtained from Abcam. Each peptide is added to Bio-Dot apparatus at 0.01 and 0.1 ug / 50uL total volume (BioRad). Let sit on rocker 60 minutes. Pull through with vacuum, wash once with 100uL TBS and twice with 200uL TBSTw. Remove membrane from Bio-Dot and transfer to 10mL Pierce SuperBlock in tray. Place on rocker for 30 minutes. Dump off SuperBlock. Added Primary Antibody in 10mL total volume. Placed on rocker for 30 minutes. Washed 3 x 5 minutes in TBSTw. Added secondary antibody diluted in SuperBlock in 10mL total volume. Placed on rocker for 30 minutes. Washed 3 x 15 minutes in TBSTw, 1 x 10 minutes in TBS. Imaged using ECL and a FluorChem Peltier-cooled camera (Cell Biosciences). The only peptide recognized by the H3K79me2 antibody was the correct one.

0.7ug unmodified Recombinant Histone H3 (Millipore 14-494) and 10ug of HepG2 nuclear lysate were resolved by electrophoresis on a 4-12% acrylamide gel. After separation, the samples were transferred to a nitrocellulose membrane with an Invitrogen iBlot system for 6 minutes. Membrane was blocked for an hour in room temperature, with 5% BSA and blotted with primary antibody diluted as indicated on the figure over night at 4C. Membrane was washed and blotted with secondary HRP-conjugated antibody. Detection was made with Optiblot ECL Detect Kit (ab133406); image was exposed for 1.5 seconds on BioRad ChemiDoc.
