- Immerse in 4 mL of blocking solution TTBS (10mM Tris HCl, pH 7.4, 0.05% Tween 20, 150mM NaCl) with 5% non-fat dried milk. Incubate on a rotator for 1 hr @ room temperature (RT). Carefully pour off blocking solution. perform a quick rinse (30 seconds) with TTBS buffer. Wash 3 x 5 minutes in TTBS. Dilute primary antibody in 4 mL of blocking solution. Add primary antibody to Array. Incubate on a rotator 1 hr @ RT. Carefully pour off blocking solution. Perform a quick rinse (30 seconds) with TTBS buffer. Wash 3 x 5 minutes in TTBS. Dilute secondary antibody in 4 mL of blocking solution. Add to array. Incubate on a rotator 1 hr @ RT. Carefully pour off blocking solution. Perform a quick rinse (30 seconds) with TTBS buffer. Wash 3 x 5 minutes in TTBS. Detect using chemi based reagents. Use BioRad ChemiDoc to expose and capture images. Graph is scaled so that saturation of signal = 1

- We performed a WB using 0.7ug unmodified recombinant Histone H3 (Millipore 14-494) and 10ug of K562 cell Lysate and 20ug of HeLa nuclear extract. Lysates were resolved by electrophoresis on a 4-12% bis-tris gel. After separation, the samples were transferred to a nitrocellulose membrane with an Invitrogen iBlot system. Membrane was blocked for an hour in room temperature, with Superblock and blotted with primary antibody in the appropriate concentration over night at 4c. Membrane was washed and blotted with secondary HRP-conjugated antibody. Detection was made with Optiblot ECL Detect Kit (ab133406) for 2 min. Band of expected size (~17kDa) visualized in HeLa nuclear extract and cannot be detected in unmodified recombinant Histone H3. Antibody seems to be specific since no reactivity with non histone proteins or unmodified histones was detected.
