- We performed a DB analysis using the large set of modified peptides provided in the Active Motif MODified Histone Peptide Array, catalog #13005. The only peptide recognized by the H3K27me3 antibody was the correct one.

- Series of Vendor validations including Western Blots, Peptide Array, ELISA. Western Blots were performed on 15 and 20ug of HeLa whole cell lysate prepared using 1% SDS hot lysis method (lane 1) and 20ug of HeLa whole cell lysate prepared using RIPA buffer. The samples were resolved by gel electrophoresis and transferred to a membrane. One membrane was blocked in 5% NFDM/TBST while the other was blocked with 2% BSA/TBST. Goat anti rabbit IGg (h&L (HRP) ab97051 (1/5000) was used as a secondary antibody. After blots were washed, Optiblot or similar reagent was used for visualization of bands. The exposure time was 40s.
