Caption
Immunoprecipitation was performed on nuclear extracts from the cell line: K562, using the antibody A303-331A. The blot shows western blot analysis of input, flowthrough, immunoprecipitate and mock immunoprecipitate using IgG.Molecular Weight: 102.023
Reviewer comment
Not readily interpretable, but looks to be multiple bands where the marked band is not >50% of the total signal in the lane.

Caption
Immunoprecipitation was performed on nuclear extracts from the cell line K562 using the antibody A303-331A. Lane 1: input nuclear lysate. Lane 2: material immunoprecipitated with antibody. Lane 3: material immunoprecipitated using control IgG. Marked bands were excised from gel and subjected to analysis by mass spectrometry. Target molecular weight: 102.023.
Submitter comment
We detected ZZZ3 in the marked immunoreactive bands by mass spec.
Reviewer comment
ZZZ3 was detected in both bands A and B in associated mass-spec.

IP followed by mass spectrometry. Briefly, protein was immunoprecipitated from K562 nuclear cell lysates using the antibody A303-331A, and the IP fraction was loaded on a 10% polyacrylamide gel (NuPAGEBis-Tris Gel) and separated with an Invitrogen NuPAGE electrophoresis system. The gel was stained by ColloidialCoomassie G-250 stain, gel fragments corresponding to the bands indicated were excised. Then proteins were trypsinized using the in-gel digestion method. Digested proteins were analyzed on an Orbitrap Elite mass spectrometer (Thermo Scientific) by the nanoLC-ESI-MS/MS technique. Peptides were identified by the SEQUEST algorithm and filtered with a high confidence threshold (Peptide false discovery rate < 1%, 2 unique peptides per protein minimum, mass error < 10 ppm).
Submitter comment
LUZP1, SUN2, C3, CSRP2BP, ALB, TTR and HSPA1A are not TFs. YEATS2 has interaction with ZZZ3, https://thebiogrid.org/117482/summary/homo-sapiens/zzz3.html.
Reviewer comment
The submitters have provided references showing interactions between ZZZ3 and YEATS2, which was ranked higher than ZZZ3 in the ranked peptide list.

ZZZ3 was detected in both bands A and B by mass spec in K562.
Reviewer comment
Marked band is not within 20% of the expected size but is consistent with the band used in mass-spec analysis in K562, which detected ZZZ3.
