Signal in IP lane for molecular weights > 17kD can be explained as background which shows in the IgG lane (lane 3). Non-reactivity of this antibody lot against unmodified H3 is demonstrated on the dot blot.

Performed dot blot analysis using Millipore’s AbSurance™ Complete Core Histone Antibody Specificity Array (cat# 16-667, http://www.millipore.com/catalogue/item/16-667#). The blot has histone H3 peptides. The antibody recognized the H3K9me3 peptide 100%, while the recombinant unmodified version of that histone shows a lack of reactivity.
