The expected size is 40kDa. In the submitted blot, the major immunoreactive band appears to be outside of the allowed 20% size deviation.
Reviewer comment
Though the major band is running higher than the expected size (>20% allowed size deviation), the accompanying knockdown secondary characterization shows that same sized band is specifically knocked down by siRNAs against YBX3.

Western blot following shRNA against YBX3 in K562 and HepG2 whole cell lysate using YBX3 specific antibody. Lane 1 is a ladder, lane 2 is K562 non-targeting control knockdown, lane 3 and 4 are two different shRNAs against YBX3. Lanes 5-8 follow the same pattern, but in HepG2. YBX3 protein appears as the green band, GAPDH serves as a control and appears in red.
