- An acid extract of Hela cells was fractionated by SDS/PAGE and transferred to a membrane for western analysis. The membrane was cut into strips and incubated with the indicated antibody, diluted at 1:500, 1:2,500, and 1:5,000 in lanes 1,2, and 3 (respectively). The preponderant staining has an apparent molecular weight consistent with migration of Histone H3. Molecular weight markers were used in lane 4 and are discernible in the image by negative staining. In addition, recombinant (unmodified) Histone H3 was gel fractionated and transferred to a membrane, and exposed to equivalent antibody dilutions (lanes 5, 6, and 7). No bands are seen, consistent with the specificity of the test antibody for histone that is tri-methylated at lysine-4.
