- HeLaS3 nuclear extract (5x10^7cells/IP) was incubated with 10ug of anti-NFYB or normal rabbit IgG (SC-2027, Santa Cruz Biotechnology) and immunoprecipitated with protein A sepharose beads. Input nuclear extract, supernatant after IP and elution fractions were analyzed by Western blotting as above, except: 10-14% gradient gel, 1:200 dilution anti-NFYB. Specific immunoprecipitation of the NFYB doublet was observed only in the anti-NFYB IP (indicated by arrows). Additional validation was carried out by observing specific ChIP-QPCR signals at known NFY bound genomic loci, and by comparison to NFYA ChIP-Seq data showing high degree of correlation between NFYA and NFYB read counts at NFY bound regions across 3 cell lines (Pearson correlation >0.7).

- Nuclear extract, of the indicated cell lines, were separated on a 10-14% gradient gel using SDS-PAGE, transferred to a PDF membrane and Western blotted using standard protocols. Primary antibody (anti-NFYB, gift from Prof. R. Mantovani) and secondary antibody (SC-2317, Santa Cruz Biotechnology) were used at a dilution of 1:2000 and 1:4000, respectively. Signal was detected using Luminol Reagent (SC-2048, Santa Cruz Biotechnology). Detection of the known NFYB doublet are indicated by arrows.
