- Analysis of Spt5 ChIP-Seq. confirms specificity of ChIP. A 34% of Spt5 ChIP-Seq peaks (p-value < 1e-10) are found within 1000bp of a RefSeq transcriptional start site (TSS), consistent with the role of Spt5 in blocking RNA polymerase II after early elongation, the majority of peaks are found downstream of the TSS. B Spt5 peaks overlap directly with cognate negative elongation factor, NELFe, and RNAPII, and downstream of the general transcription factors TFIIB and TBP. C DSIF (Spt4/Spt5) is known to act proximal to the transcriptional start site and travel with elongating RNAPII. Consistent with a role downstream of the start site a significant number of Spt5 ChIP-Seq. peaks are found across RefSeq genes and proximal to the transcriptional end site, compared to NELFe which associates specifically with transcriptional start sites.
