- Analysis of TFIIB ChIP-Seq. confirms specificity of TFIIB ChIP. A The 66% of MACS called TFIIB ChIP-Seq peaks (p-value < 1e-10) were found within 500bp of a RefSeq TSS. The median peak position was -16 bp relative to the TSS, consistent with the role of TFIIB in recruitment of the RNA polymerase II complex. B Comparison of TFIIB MACS ChIP-Seq peaks to RNAPII, TBP, NELFe and Spt5 ChIP-Seq peaks place TFIIB coincident with TBP and downstream of the paused RNAPII and negative elongation factors. C De novo motif discovery with the MEME suite of analysis software using 100bp of sequence flanking the top 1000 TFIIB peaks identifies the TATA box as an enriched DNA element (p-value < 3e-4). The majority of TATA motifs are found within 50 bp of the TFIIB peak summit.
