- ChIP-seq was performed using a ELK4 antibody in Hela cells. The top 1000 peaks were analyzed with a de novo motif analysis (http://motif.bmi.ohio-state.edu/ChIPMotifs/). The top motif that was identified was CTTCCGG (P value: 1.0215e-318), which in the reverse direction is CCGGAAG. A previously identified motif for ELK4 is ACTTCCGGT (EMBO J. 2010 July 7; 29(13): 2147-2160), which contains the same core sequence as the de novo motif identified for ELK4. We then analyzed the percentage of peaks containing the de novo ELK4 motif in the entire set of ELK4 called peaks. We found that 88% of the top 1000 peaks contained this motif, with the percentage dropping to 75% in the top 3715 peaks the dataset.

- ELK4 Western blotting. Nuclear extracts were prepared from Hela and HEK293 cells and 25ug were loaded onto an 10% SDS-PAGE for Western blotting using anti-ELK4 antibody at a dilution of 1:1,000. A specific band corresponding to the predicted molecular weight (approximately 47 kDa) was detected and no other non-specific bands were observed.
