Caption
IP followed by mass spectrometry. Briefly, protein was immunoprecipitated from K562 nuclear cell lysates using the antibody A303-265A, and the IP fraction was loaded on a 10% polyacrylamide gel (NuPAGEBis-Tris Gel) and separated with an Invitrogen NuPAGE electrophoresis system. The gel was stained by ColloidialCoomassie G-250 stain, gel fragments corresponding to the bands indicated were excised. Then proteins were trypsinized using the in-gel digestion method. Digested proteins were analyzed on an Orbitrap Elite mass spectrometer (Thermo Scientific) by the nanoLC-ESI-MS/MS technique. Peptides were identified by the SEQUEST algorithm and filtered with a high confidence threshold (Peptide false discovery rate < 1%, 2 unique peptides per protein minimum, mass error < 10 ppm).

CHD4 and HDAC1 (55KD), HDAC2 (55KD) are in one complex, ZNF217 and HDAC1, HDAC2 are components of BHC complex, CHD4 are also part of this complex, http://www.genecards.org/cgi-bin/carddisp.pl?gene=HDAC2&keywords=hdac2 and http://www.genecards.org/cgi-bin/carddisp.pl?gene=HDAC1&keywords=hdac1. HDAC1 and HDAC2 are not in the same gel slice with CHD4 and ZNF217. Both HNRNPUL1 and ZNF217 have interactions with ELAVL1 (36KD), http://thebiogrid.org/113547/summary/homo-sapiens/znf217.html and http://thebiogrid.org/116281/summary/homo-sapiens/hnrnpul1.html. ELAVL1 is not in the same gel slice with HNRNPUL1 and ZNF217. Although chromatin-associated protein CHD4 is ranked higher by enrichment than ZNF217, the submitters note that CHD4 and ZNF217 may be part of the same complex.
